Strategies for the asymmetric functionalization of indoles: an update.

چکیده
During the past four years, the research of new synthetic methodologies for the rapid construction of enantiomerically pure substituted indole has had a fruitful and important growth. This research line continues to produce stunning arrays of enantioselective technologies either metal or organocatalyzed. Thus, an update of our previous review (Chem. Soc. Rev., 2010, 39, 4449-4465) has become necessary and this critical review documents the literature on this topic, until the end of 2013.

An easily available iron catalyst was developed to accomplish the C H functionalization of indoles with a-aryl-a-diazoesters in high yields under mild conditions. The asymmetric synthesis of 3-indolylglycine derivatives was achieved by an oxidative heterocoupling reaction. This method for the selective C-3 functionalization of unprotected indoles with the chiral equivalent of a nucleophilic glycine nickel(II) complex afforded adducts with high diastereoselectivities. The decomposition of adducts readily afforded 3-indolylglycine derivatives in high yields.
